چکیده

A fructan exohydrolase selective for (2----1)-linked terminal fructosyl linkages, isolated from barley (Hordeum vulgare L. cv. Morex) stems and leaf sheaths, was used to elucidate the chemical structures of several oligomeric fructans extracted from liliaceous and graminaceous species. Products released by enzymic and mild acid hydrolysis were separated by reversed-phase high-performance liquid chromatography. Gas-liquid chromatography-mass spectrometry of partially methylated alditol acetates permitted unequivocal deduction of many linkage sequences, first of the hydrolysis products and then of the original oligomers. We found that bifurcose, a tetrasaccharide formed by addition of a fructosyl unit to O-6 of the central fructose residue of 1-kestose, was a central molecule in the generation of the branched, oligomeric fructans of wheat (Triticum aestivum L. cv. Fidel). These arise by the extension of both (2----1)- and (2----6)-linked chains from the bifurcose branch-point residue. Some of the (2----6)-linked units that slowly accumulate in oligomers may arise in vivo from selective hydrolysis, by fructan exohydrolases, of (2----1)-linked terminal units at branch point residues rather than by the action of (2----6)-specific synthases. Limited hydrolysis by specific exohydrolases in vitro coupled with separation of the oligomeric products constitutes an effective approach to the sequence analysis of complex oligosaccharides.

Defoliation induces fructan 1-exohydrolase II in Witloof chicory roots. Cloning and purification of two isoforms, fructan 1-exohydrolase IIa and fructan 1-exohydrolase IIb. Mass fingerprint of the fructan 1-exohydrolase II enzymes.

The cloning of two highly homologous chicory (Cichorium intybus var. foliosum cv Flash) fructan 1-exohydrolase cDNAs (1-FEH IIa and 1-FEH IIb) is described. Both isoenzymes could be purified from forced chicory roots as well as from the etiolated "Belgian endive" leaves where the 1-FEH IIa isoform is present in higher concentrations. Full-length cDNAs were obtained by a combination of reverse t...

Fructan 1-exohydrolases. beta-(2,1)-trimmers during graminan biosynthesis in stems of wheat? Purification, characterization, mass mapping, and cloning of two fructan 1-exohydrolase isoforms.

Graminan-type fructans are temporarily stored in wheat (Triticum aestivum) stems. Two phases can be distinguished: a phase of fructan biosynthesis (green stems) followed by a breakdown phase (stems turning yellow). So far, no plant fructan exohydrolase enzymes have been cloned from a monocotyledonous species. Here, we report on the cloning, purification, and characterization of two fructan 1-ex...

Stem carbohydrate dynamics and expression of genes involved in fructan accumulation and remobilization during grain growth in wheat (Triticum aestivum L.) genotypes with contrasting tolerance to water stress

The genetic and physiological mechanisms underlying the relationship between water-soluble carbohydrates (WSC) and water stress tolerance are scarcely known. This study aimed to evaluate the main WSC in stems, and the expression of genes involved in fructan metabolism in wheat genotypes growing in a glasshouse with water stress (WS; 50% field capacity from heading) and full irrigation (FI; 100%...

Activities of fructan- metabolizing enzymes in barley stems subjected to terminal drought stress

Barley crop grown in semiarid areas may experience water deficit especially during grain filling that makes them more dependent on stem water-soluble carbohydrates (WSC). Fructans are the most important reserved carbohydrates. A pot experiment was undertaken at Shahid Chamran university in the duration of 2010-2011 growing seasons to investigate the accumulation and loss of WSC and the activity...

Localization of the Enzymes of Fructan Metabolism in Vacuoles Isolated by a Mechanical Method from Tubers of Jerusalem Artichoke (Helianthus tuberosus L.).

Vacuoles isolated by a mechanical slicing method from developing tubers of Jerusalem artichoke (Helianthus tuberosus L.) contain activities of the two principal enzymes responsible for fructan synthesis: sucrose-sucrose fructosyl transferase and fructan-fructan fructosyl transferase. Both enzymes are associated with the vacuolar sap and not with the tonoplast. In vacuoles isolated from dormant ...
